Studies in cryptococcal fever. I. Responses to intact organisms and to a soluble agent derived from cryptococci.

چکیده
It is a long-established clinical observation that most microbial infections are associated with fever. With many of these agents, however, the pathogenesis of fever has been only recently elucidated and it is apparent that several different mechanisms may be implicated.' The following studies were undertaken to compare fevers induced in rabbits by a nonpathogenic fungus, Cryptococcus albidus, with those reported in similar models employing viruses' and both Gram-negative' and Gram-positive bacteria."In contrast to extensive investigations with other microbes, there are only a few previous studies demonstrating the pyrogenicity of various fungi." Both the whole cells and soluble products of these microorganisms have been utilized to produce fever, but hitherto no systematic attempt has been made to determine if different pyrogenic mechanisms were involved; nor has there been any previous effort to establish the relationship of fevers induced by fungi to those caused by bacterial pyrogens, the co-called "endotoxins" of Gram-negative bacteria. In the following studies, evidence is presented that both living and dead cells of this cryptococcal species are pyrogenic in normal rabbits, whereas its soluble products seem to cause fever only, or primarily, in specifically sensitized animals. Further, in their pyrogenic actions, the cells and soluble products of this fungus appear to be clearly separable from each other, as well as from Gram-negative bacterial endotoxins, and in this regard resemble fever-inducing factors associated with the cells and culture filtrates, respectively, of certain phage types of staphylococci.1-17

Studies in cryptococcal fever. II. Responses of sensitized and unsensitized rabbits to various substances derived from cryptococcal cells.
The cause of fevers induced in rabbits by intravenous injection of cryptococcal cells and soluble extracts was investigated in a previous paper in this series.' Two distinct types of pyrogenic responses were found to be produced by this organism. Intact cells, whether live or killed, caused fevers in normal rabbits, whereas the soluble material induced responses primarily in animals that had be...
متن کاملAge-related resistance of C57BL/6 mice to Cryptococcus neoformans is dependent on maturation of NKT cells.
Conflicting results have been reported regarding the ability of C57BL/6 mice to clear infections due to Cryptococcus neoformans. Examination of the various experimental protocols used suggested that C57BL/6 mice might develop the ability to resist infection as they mature. We analyzed the ability of C57BL/6 mice of different ages to respond to immunization with cryptococcal antigen or to clear ...
متن کاملA Rigorous Calculation Method for Determining Potential-pH Diagrams Part I: Copper in Aqueous Solutions of Various Complexing Agents
The main aim of this research is to determine optimal leaching conditions; Eh, pH and complexing agent concentrations, for recovery of Copper from hydroxide sludges such as those produced by electroplating shops, metal finishers, treatment of acid mine drainage, and industrial wastewater in general. This has been preceded by a theoretical approach and numerical and computer calculation. Pot...
متن کاملOpsonization of encapsulated Cryptococcus neoformans by specific anticapsular antibody.
Antisera prepared in rabbits against either whole encapsulated cells of Cryptococcus neoformans or purified cryptococcal polysaccharide were opsonic for the encapsulated yeast. The opsonic activity was removed by absorption with whole cryptococci and was inhibited by free polysaccharide. As little as 0.13 microgram of cryptococcal polysaccharide produced a 50% inhibition of opsonization. Variou...

In vitro lymphocyte responses of high responder (HR) and low responder (LR) guinea pigs from pe-ripheral blood lymphocytes (PBL) to parasite antigens soluble protein third stage larvae (SPL3) and soluble protein adult stage ( SPA), non-parasite antigen ovalbumin (OVA) were examined. There was substantial differences between HR and LR guinea pigs in the rate of acquisition of responsiveness to t...
